The involvement of astrocytes in the progression of Alzheimer’s disease

Knowledge about the cellular mechanisms behind the propagation of Alzheimer’s disease (AD) in the brain is limited. Decades of research have focused on neuronal abnormalities in AD, but recently more attention has been given to glial cells. The aim of this project is to clarify the involvement of astrocytes in the progression of AD and to investigate their therapeutic potential. Our previous results demonstrate that astrocytes engulf large amounts of aggregated amyloid-beta that is stored, rather than degraded by the cells. The accumulation results in lysosomal defects and in the secretion of extracellular vesicles with neurotoxic content. Our hypothesis is that astrocytes try to be ‚Äúhelpful‚Äù by ingesting amyloid-beta, but are overwhelmed by the challenge and instead promote disease spreading and neuronal cell death. To elucidate the impact of astrocytes in the propagation of amyloid-beta pathology, we will perform cell culture experiments, in vivo studies and analyze brain tissue from AD patients. We will examine whether astrocytes spread amyloid-beta predominately via secretion or via tunneling nanotubes and identify strategies that reduce the spreading. Moreover, we will investigate the potential of transplanted astrocytes, containing amyloid-beta deposits, to induce pathology in an AD mouse model. Being the most numerous glial cell type in the brain, astrocytes constitute a compelling treatment target and our results will contribute important data to the research field.
