SMALL CELL LUNG CANCER DEVELOPMENT BY YAP1 USING MICE AS A MODEL

Abstract [en]

Various combinations of surgery followed up by chemotherapy is today the standard treatment for patients with lung cancer, considering the fact that the survival rate of patients with lung cancer is also low, early detection and treatment are essential to establish. YAP1 has been described as an oncogene, involved in the Salvador-Warts-Hippo pathway which regulates apoptosis, cell differentiation and proliferation, although another study suggest Yes - Associated Protein 1 (YAP 1) as a tumour suppressor. The purpose of this study was to introduce YAP1 into a triple knockout model, estimate tumour development of primary and secondary transplanted mouse Small Cell Lung Cancer (SCLC). Estimation of tumour volume from transplanted mSCLC and MTC, staining of secondary tumour tissue and western blot of cultured cells were the methods used in this study. Transplanted mSCLC`s showed similar pattern to cultured cells, even though the injected MTC cells developed in a different speed compared to the transplanted cells. Staining of the tumour tissue confirms the irregular structure and colour of the secondary tumour cells observed. The cultured mSCLC also expressed YAP1, though not the secondary tumour cells. Using transplantation opposed to culturing of cells as a method to develop tumours, have shown some differences between Medullary Thyroid Cancer cells, also YAP1 were only expressed by the primary cells using western blot. Maintaining the introduction of YAP1 into the new quadruple knockout model, will perhaps give a wider perspective on the function but also interaction of YAP1 as oncogene or tumour suppressor.
