COMPARISON OF THE CRYOPROTECTANT DMSO FROM TWODIFFERENT SUPPLIERS

Abstract [en]

Cryopreservation is a method that allows long-term storage of cells by preservingthe cells at extremely low temperatures. The method is used in various medicalfields, such as stem cell transplantation and fertility treatment. To protect the cellsduring freezing, different cryoprotectants are used. These minimize the risk of icecrystal formation, which is the most common cause of cell death during cryopreservation.The aim of this study was to evaluate and compare DMSO, a cryoprotectant, fromtwo suppliers.In this study, 10 blood samples were used at the Uppsala University Hospital.Leukocytes were isolated from whole blood by using ficoll separation. The leukocyteswere frozen using freezing medium containing either one of two differentDMSO, and thereafter thawed. Cell viability was evaluated by use of microscopyand flow cytometry. In addition, cell functionality was tested using phytohemagglutinin(PHA) stimulation and subsequent measurement of ATP.For the results of this study, a paired samples t-test average on a 95% significancelevel was used where t < 1.833. For microscopy obtained a t-value of 0.15 andt-value for flowcytometri was 0.19. For Immuknow test obtained a t value of 0.97.The results indicates that there was no significance difference between DMSO fromthe two different suppliers.
