Abstract [en]

Subcutaneous injection is a commonly used route of drug administration for both small molecules and biologics. In order to facilitate the development of new subcutaneously injected drugs, models that can predict the pharmacokinetic profile of subcutaneously injected drugs are in great demand. PK-sim® (Bayer Technology Service GmbH, Germany) is an open-source PBPK software tool that allows whole-body PBPK modelling for humans and common laboratory animals. The study aimed to identify whether PK-sim® can be used to predict the plasma profiles of drugs administered subcutaneously. Prediction of a total selection of fourteen drugs specifically with differing physicochemical properties were developed by PK-sim®: eight for small molecules and six for biologics. Most drugs were described with three species: human, non-rodent, and rodent. PK-sim® could not predict absorption phase for small molecules and showed poor performance in both small molecules and biologics.
