    Abstract [en]

    The research field of systems biology has gained a lot of interest during the last decades. Systems biology can be seen as the systematic study of complex interactions in biological systems, mainly by methods from dynamical systems theory. This thesis mainly deals with the testosterone (Te) regulation in the human male, but techniques developed here might be useful for studying other parts of the endocrine system too. The contribution of the thesis can be divided into two parts: one covering mathematical models of Te regulation and another suggesting and validating identification techniques for those models. Regarding modeling, existing models of testosterone regulation have been extended with time delays and nonlinear dynamics, with the purpose of achieving better fit to clinical data. The identification part treats the estimation of unknown model parameters, and estimation of signals that cannot be measured in a noninvasive way.

    The continuous part of a hybrid (pulse-modulated) model of testosterone (Te) feedback regulation in the human male is extended with infinite-dimensional and nonlinear blocks, to obtain the dynamics that better agree with the hormone concentration profiles observed in clinical data. A linear least-squares based optimization algorithm is developed for the purpose of detecting impulses of gonadotropin-releasing hormone (GnRH) from measured concentration of luteinizing hormone (LH). The estimated impulse parameters are instrumental in evaluating the frequency and amplitude modulation functions parameterizing the pulse-modulated feedback. The proposed approach allows for the identification of all model parameters from the hormone concentrations of Te and LH. Simulation results of the complete estimated closed-loop system exhibiting similar to the clinical data behavior are provided.

    Abstract [en]

    Dynamical behaviors arising due to continuous exogenous signals in a previously developed pulse-modulated mathematical model of non-basal testosterone regulation in the human male are studied. In the context of endocrine regulation, exogenous signals represent e.g. the drugs used in hormone replacement therapies, the effects due to circadian rhythm, and interactions with other endocrine loops of the organism. The model dynamics are hybrid since the hormone metabolism is suitably captured by a continuous description and the control feedback is implemented in a discrete (i.e. event-based) manner by the hypothalamus of the brain. It is demonstrated that the endocrine loop with an exogenous signal entering the continuous part can be equivalently described by proper modifications in the pulse-modulation functions of the autonomous model. This extends the scope of the autonomous pulse-modulated models of endocrine regulation to a broader class of problems, such as therapy optimization, where an exogenous signal is involved. The cases of constant and harmonic exogenous signals are treated in detail and illustrated by bifurcation analysis.
