    Abstract [en]

    We present an amplitude analysis of the decay D-0 -> K- pi(+)pi(+)pi(-) based on a data sample of 2.93 fb(-1) acquired by the BESIII detector at the psi(3770) resonance. With a nearly background free sample of about 16000 events, we investigate the substructure of the decay and determine the relative fractions and the phases among the different intermediate processes. Our amplitude model includes the two-body decays D-0 -> (K) over bar*(0)rho(0), D-0 -> K- a(1)(+) (1260) and D-0 -> K-1(-)(1270)pi(+), the three-body decays D-0 -> K-1(-)*(0)pi(+)pi(-) and D-0 -> K- pi(+)rho(0), as well as the four-body nonresonant decay D-0 -> K- pi(+)pi(+)pi(-). The dominant intermediate process is D-0 -> K(-)a(1)(+)(1260)accounting for a fit fraction of 54.6%.

    Studies of Be migration in the JET tokamak using AMS with Be-10 marker2016Konferansepaper (Fagfellevurdert)
    Abstract [en]

    The JET tokamak is operated with beryllium limiter tiles in the main chamber and tungsten coated carbon fiber composite tiles and solid W tiles in the divertor. One important issue is how wall materials are migrating during plasma operation. To study beryllium redistribution in the main chamber and in the divertor, a Be-10 enriched limiter tile was installed prior to plasma operations in 2011-2012. Methods to take surface samples have been developed, an abrasive method for bulk Be tiles in the main chamber, which permits reuse of the tiles, and leaching with hot HCl to remove all Be deposited at W coated surfaces in the divertor. Quantitative analysis of the total amount of Be in cm(2) sized samples was made with inductively coupled plasma atomic emission spectroscopy (ICP-AES). The Be-10/Be-9 ratio in the samples was measured with accelerator mass spectrometry (AMS). The experimental setup and methods are described in detail, including sample preparation, measures to eliminate contributions in AMS from the B-10 isobar, possible activation due to plasma generated neutrons and effects of diffusive isotope mixing. For the first time marker concentrations are measured in the divertor deposits. They are in the range 0.4-1.2% of the source concentration, with moderate poloidal variation.

    Abstract [en]

    The rate of the reaction between iodide and selenium(IV) at trace levels to form selenium and iodine has been determined in 1–6M hydrochloric acid. The reaction rate increases rapidly with acidity. When hydrochloric acid is added to reduce selenate to selenite prior to the determination of total selenium, some selenium may be lost by reduction to the element if iodide is present. A table of half-lives of the selenite—iodide reaction under various conditions is presented. A method for removal of iodide is suggested.

    Abstract [en]

    Column-switching liquid chromatography followed by low resolution ICP-MS was evaluated as a tool for speciation analysis of aluminium-containing biomolecules. The strategy was applied on siderophores, small organic molecules (Mr < 1500) which normally act as strong iron chelators. The drawbacks normally encounterd with aluminium detection using low resolution ICP-MS are the formation of polyatomic ions causing isobaric overlaps and space-charge effects. When adding a carbon rich solvent, such as methanol or acetonitrile, the 13C14N+, 12C15N+ and 12C14N1H+ with the same mass as 27Al+ will form in the plasma. The nitrogen is either entrained from the surrounding atmosphere or added with the constituents in the mobile phase. These disadvantages were successfully counteracted by the use of nitrogen free organic modifier in the mobile phase and the use of cool plasma conditions. Detection limits for standard solutions of aluminium-chelated ferrichrome in sub-nanomolar range were obtained by monitoring the aluminium-27 isotope. The combined use of LC-ICP-MS and LC-ESI-MS/MS was also evaluated as a tool to identify unknown metal complexes, here siderophores, in field soil solution samples. Two aluminium-chelated siderophores, Al-desferrichrom and Al-desferricrocin, were identified and quantified. Both aluminium-siderophore complexes were present in the low nanomolar range (1.1 and 0.7 nM, respectively).

    In this study, we used proteomics in conjunction with microscopy to study differences in the proteome and hyphal morphology of the copper tolerant soft rot fungus Phialophora malorum grown in media containing 0.064, 0.64% Cu as CuSO4. Unique proteins were found in the control and the copper-treated (0.064% CuSO4) samples. Of five unique proteins found in the 0.064% CuSO4 treated cultures, ATP synthase subunit alpha is considered to play an important role in copper tolerance as it is involved in the biosynthesis of fatty acids and steroids and may relate to morphological changes associated with hyphal cell walls of the fungus when grown in the presence of copper. ICP-AES analyses showed total mycelial Cu to increase with media Cu with 5246- and 16535 mu g Cu/g dry wt mycelia respectively found in 0.064 and 0.64% Cu-cultures after 6 weeks growth. Rubeanic acid staining of 0.064% mycelia showed Cu bound in intracellular bodies while most Cu was found as extracellular precipitates on the surfaces of hyphae in 0.64% Cu. SEM showed hyphal surfaces enrobed in fibrillar polysaccharides to which Cu was bound.

    ICP-MS is a sensitive element analysis technique used for analyzing several different sample types. This can result in difficult matrixes which can affect both physical parameters and create overlaps of analyte elements. Some of the possible overlaps can be reduced by the use of reaction and/or collision cells, while the use of internal standards can help with reducing the physical interferences caused by a matrix. While both internal standardization and the use of cells have been studied separately, their effects on each other have not been investigated earlier. In this study ICP-MS was used to analyze Mg-24, Al-27, Ti-47, Ti-49, V-51, Cr-52, Cr-53, Mn-55, Fe-57, Co-59, Ni-60, Ni-61, Ni-62, Cu-63, Cu-65, Zn-66, Zn-67, As-75, Se-78, Se-82, Cd-111, and Pb-208 with Be-9, Y-89, Ga-69, Rh-103, In-115, Ir-193, and Tl-205 as internal standards with high concentrations of either HNO3, PBS-buffer, or Triton X-100 as the matrix, in reaction-, collision- and standard-cell modes. This was done to investigate which internal standards would compensate matrix effects in different cell modes. All internal standards, except Be, compensated fairly well (relative sensitivity RSD < 10%) even for severe matrix effects for most elements regardless of similarity in mass in the different cell modes. For Zn, As and Se no proper internal standard could be found, of the ones investigated.

    Electrochemically controlled solid-phase extractions of anions were interfaced on-line to electrospray mass spectrometry (ESI-MS) and inductively coupled plasma mass spectrometry (ICP-MS), using polypyrrole coated electrodes and a thin-layer electrochemical (EC) flow cell. The results indicate that electrochemically controlled solid-phase extraction (EC-SPE) can be used as a versatile potential controlled sample preparation technique for a range of anions and that the properties of the polypyrrole coatings can be modified by altering the electrodeposition conditions. In the present study, the influence of interfering anions (i.e., fluoride and sulfate), and the anion used during the electropolymerisation, on the bromide extraction recovery was investigated for EC-SPE interfaced to ICP-MS. The results of these experiments show that the interference due to the presence of similar concentrations of sulfate can be reduced when using a polypyrrole coating electropolymerised in the presence of bromide ions. Scanning electron microscopy (SEM) and atomic force microscopy (AFM) measurements were also used to study the morphology of the coatings, as well as the variations in the film thickness within the coatings. The effect of different desorption techniques on the bromide preconcentration factor in the ICP-MS on-line flow system was also examined. Stopped-flow desorption was found to give rise to significantly increased preconcentration factors in comparison with desorptions in flowing solutions. While the desorption efficiency depends on the type of desorption electrolyte (the electrolyte in which the desorption takes place), due to the competing influx of cations, the influence of the pH on the switching charge of the polypyrrole coating was found to be small, at constant ionic strength. To study the applicability of the EC-SPE technique with respect to real samples, investigations were also made with tap water samples spiked with different bromide concentrations. The results of these experiments, which were carried out using a modified thin-layer EC flow cell allowing in situ polymerisation of polypyrrole yielding a polymer plug covering the cross section of the channel, demonstrate that 3 uM concentrations of bromide could be detected in the tap water sample. This demonstrates that the extraction technique allows extractions of low concentrations of ions in the presence of significantly higher concentrations of other similar ions. The fact that the extraction and desorption steps are electrochemically controlled makes EC-SPE particularly well suited for inclusion in miniaturised lab-on-a-chip systems.

    While the use of silicon‐based electrodes can increase the capacity of Li‐ion batteries considerably, their application is associated with significant capacity losses. In this work, the influences of solid electrolyte interphase (SEI) formation, volume expansion, and lithium trapping are evaluated for two different electrochemical cycling schemes using lithium‐metal half‐cells containing silicon nanoparticle–based composite electrodes. Lithium trapping, caused by incomplete delithiation, is demonstrated to be the main reason for the capacity loss while SEI formation and dissolution affect the accumulated capacity loss due to a decreased coulombic efficiency. The capacity losses can be explained by the increasing lithium concentration in the electrode causing a decreasing lithiation potential and the lithiation cut‐off limit being reached faster. A lithium‐to‐silicon atomic ratio of 3.28 is found for a silicon electrode after 650 cycles using 1200 mAhg−1 capacity limited cycling. The results further show that the lithiation step is the capacity‐limiting step and that the capacity losses can be minimized by increasing the efficiency of the delithiation step via the inclusion of constant voltage delithiation steps. Lithium trapping due to incomplete delithiation consequently constitutes a very important capacity loss phenomenon for silicon composite electrodes.

    Column-switching liquid chromatography followed by low-resolution ICPMS was evaluated as a tool for speciation analysis of metal-containing biomolecules. The strategy was applied on siderophores, strong iron chelators of low molecular weight (M(w) < 1500). Prior to the LC-ICPMS analysis, reductive displacement of iron by gallium was performed using ascorbate as the reducing agent to increase the sensitivity. Different experimental conditions during the exchange reaction were tested using ferrichrysin and ferrichrome for evaluation. A reaction time of 30 min and a pH of 3.9 gave an exchange yield of 27 and 83% for ferrichrysin and ferrichrome, respectively. A gradient elution profile was also developed to separate gallium-chelated siderophores on a PGC column. Detection limits for standard solutions of ferrichrysin and ferrichrome in the low-nanomolar range were obtained by monitoring the gallium-69 isotope. The combined use of LC-ICPMS and LC-ESI-MS/MS was also evaluated as a tool to identify unknown metal complexes, here siderophores, in field soil solution samples.

    PURPOSE:

    Perfusion assessment by monitoring the transport of a tracer bolus depends critically on conversion of signal intensity into tracer concentration. Two main assumptions are generally applied for this conversion; (1) contrast agent relaxivity is identical in blood and tissue, (2) change in signal intensity depends only on the primary relaxation effect. The purpose of the study was to assess the validity and influence of these assumptions.

    MATERIALS AND METHODS:

    Blood and cerebral tissue relaxivities r1, r2, and r2* for gadodiamide were measured in four pigs at 1.5 T. Gadolinium concentration was determined by inductively coupled plasma atomic emission spectroscopy. Influence of the relaxivities, secondary relaxation effects and choice of singular value decomposition (SVD) regularization threshold was studied by simulations.

    RESULTS:

    In vivo relaxivities relative to blood concentration [in s-1 mM-1 for blood, gray matter (GM), white matter (WM)] were for r1 (2.614 ± 1.061, 0.010 ± 0.001, 0.004 ± 0.002), r2 (5.088 ± 0.952, 0.091 ± 0.008, 0.059 ± 0.014), and r2* (13.292 ± 3.928, 1.696 ± 0.157, 0.910 ± 0.139). Although substantial, by a nonparametric test for paired samples, the differences were not statistically significant. The GM to WM blood volume ratio was estimated to 2.6 ± 0.9 by r1, 1.6 ± 0.3 by r2, and 1.9 ± 0.2 by r2*. Secondary relaxation was found to reduce the tissue blood flow, as did the SVD regularization threshold.

    CONCLUSION:

    Contrast agent relaxivity is not identical in blood and tissue leading to substantial errors. Further errors are introduced by secondary relaxation effects and the SVD regularization.

    PURPOSE: To study the uptake, toxicity and radiation effects in vitro of a diol-amino acid-carborane (DAAC-1) and make comparisons with the previously studied diol-amine-carborane (DAC-1). MATERIALS AND METHODS: Toxicity and radiation effects were studied with clonogenic survival, uptake by measuring the cellular boron content and the subcellular distribution was investigated after organelle separation with centrifugation. The studied cell line was human glioma U343. RESULTS: DAAC-1 showed an accumulation of 1-1.5 times, compared with the culture medium, and was non-toxic up to 47 microg boron/ml. The accumulation of DAC-1 was about 90 times, but toxic effects were detectable already at the concentration 5 microg boron/ml. None of the compounds was localized in the cell nucleus. Following irradiation with thermal neutrons, DAC-1 was about 2.5 times more effective than DAAC-1 and about 4.9 times more effective than neutrons alone, at the survival level 0.2. The dose modifying factors, when compared with the neutron beam alone, were for both DAAC-1 and DAC-1 about 1.5 and about 5 when compared with 60Co-gamma-radiation. CONCLUSIONS: DAAC-1 was less toxic than DAC-1 but gave less accumulation of boron. Both substances gave significant boron-dependent cell inactivation when the test cells were exposed to thermal neutrons.

    Abstract [en]

    A method for the detection and speciation of inositol phosphates (InsP(n)) in sediment samples was tested, utilizing oxalateoxalic acid extraction followed by determination by high-performance liquid chromatography coupled with tandem mass spectrometry (HPLCMS/MS) using electrospray ionization (ESI) in negative mode. The chromatographic separation was carried out using water and ammonium bicarbonate as mobile phase in gradient mode. Data acquisition under MS/MS was attained by multiple reaction monitoring. The technique provided a sensitive and selective detection of InsP(n) in sediment samples. Several forms of InsPn in the oxalateoxalic acid extracted sediment were identified. InsP(6) was the dominating form constituting 0.250 mg P/g DW (dry weight); InsP(5) and InsP(4) constituted 0.045 and 0.014 mg P/g DW, respectively. The detection limit of the LCESI-MS/MS method was 0.03 mu M InsPn, which is superior to the currently used method for the identification of InsPn, P-31 nuclear magnetic resonance spectroscopy (P-31 NMR). Additionally sample handling time was significantly reduced.
