Knowledge and beliefs about HPV and HPV vaccine among young Thai females2016Independent thesis Basic level (degree of Bachelor), 10 credits / 15 HE creditsStudent thesis
    Abstract [en]

Background: Human Papilloma Virus (HPV) is the recognized main reason for developing cervical cancer. HPV vaccine given to females is the most effective prevention. Purpose: To investigate knowledge and beliefs about HPV, cervical cancer and HPV vaccine among young Thai females in north-eastern part of Thailand. Further, to discover potential differences between those stating having knowledge about HPV and cervical cancer (group SHK), and those stating not having knowledge about HPV and cervical cancer (group SNHK). Method: A cross-sectional survey using a questionnaire about knowledge and beliefs of HPV where 221 young Thai females, aged 18-21, participated. Orem’s self-care theory was used as theoretical framework. Result: Less than 50 % of the participants knew about visible signs and symptoms of HPV infection. However, over 70 % had knowledge regarding HPV´s relation to sexual activity. Internet was the greatest source of information about HPV. Participants had positive belief towards the vaccine and more than 95% wished to get vaccinated. Group SHK had more knowledge then group SNHK with significant difference in seven out of fourteen knowledge items, and showed more positive beliefs with significant difference in six out of sixteen belief statements. Conclusion: The overall level of knowledge about HPV and cervical cancer was insufficient. However, this did not affect the participant’s beliefs in the subject negatively. Health care should provide viable internet sites with information about HPV to ensure that young Thai females get requisites, enabling self-care on preventing HPV infections by vaccination.

The Role of Polyadenylation in Human Papillomavirus Type 16 Late Gene Expression2005Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

High-risk type human papillomaviruses (HPVs) are associated with cancer. HPVs are strictly epitheliotropic and infect basal cell layers, establishing a life cycle strongly linked to the differentiation stage of the infected cells. The viral capsid late genes, L2 and L1, are only expressed in terminally differentiated epithelium. Late gene expression involves regulation of most gene processing events including transcription, splicing, polyadenylation, mRNA stability and translation.

Both L2 and L1 have elements present in the open reading frames (ORFs) negatively affecting mRNA levels and translation. The negative elements in L1 were mapped to the first 514 nucleotides, with the strongest inhibitory effect located in the first 129 nucleotides. The negative elements in the L2 sequence were concentrated in two locations on the gene. Both genes were mutated by changing the nucleotide sequence while retaining the amino acid sequence. Mutating the first 514 nucleotides in L1 deactivated the negative elements while the entire L2 gene had to be mutated to achieve the same result. The L2 protein was found to localise the L1 protein into a punctuated pattern in the nucleus.

In the HPV-16 genome the negative elements reside in regions important for regulation of polyadenylation and splicing, critical for late gene expression. By exchanging parts of the L2 gene in subgenomic constructs with the corresponding mutant sequence we show that certain features of the L2 elements direct splicing to the L1 splice acceptor, and also regulate the efficiency of the early polyadenylation site. Cumulative binding of hnRNP H to the L2 mRNA gradually increased polyadenylation efficiency. Most interestingly, hnRNP H levels were downregulated in more differentiated epithelial cells.

Elucidation of how expression of the immunogenic late proteins is regulated would be greatly beneficial in prevention and treatment of HPV infection and thereby cancer.

    Abstract [en]

Production of human papillomavirus type 16 (HPV-16) virus particles is totally dependent on the differentiation-dependent induction of viral L1 and L2 late gene expression. The early polyadenylation signal in HPV-16 plays a major role in the switch from the early to the late, productive stage of the viral life cycle. Here, we show that the L2 coding region of HPV-16 contains RNA elements that are necessary for polyadenylation at the early polyadenylation signal. Consecutive mutations in six GGG motifs located 174 nucleotides downstream of the polyadenylation signal resulted in a gradual decrease in polyadenylation at the early polyadenylation signal. This caused read-through into the late region, followed by production of the late mRNAs encoding L1 and L2. Binding of hnRNP H to the various triple-G mutants correlated with functional activity of the HPV-16 early polyadenylation signal. In addition, the polyadenylation factor CStF-64 was also found to interact specifically with the region in L2 located 174 nucleotides downstream of the early polyadenylation signal. Staining of cervix epithelium with anti-hnRNP H-specific antiserum revealed high expression levels of hnRNP H in the lower layers of cervical epithelium and a loss of hnRNP H production in the superficial layers, supporting a model in which a differentiation-dependent down regulation of hnRNP H causes a decrease in HPV-16 early polyadenylation and an induction of late gene expression.

Improved potency and selectivity of an oncolytic E1ACR2 and E1B19K deleted adenoviral mutant in prostate and pancreatic cancers.2010In: Clinical Cancer Research, ISSN 1078-0432, E-ISSN 1557-3265, Vol. 16, no 2, p. 541-53Article in journal (Refereed)
    Abstract [en]

PURPOSE Replication-selective oncolytic adenoviruses are a promising class of tumor-targeting agents with proven safety in hundreds of patients. However, clinical responses have been limited and viral mutants with higher potency are needed. Here, we report on the generation of a novel set of mutants with improved efficacy in prostate and pancreatic carcinoma models. Currently, no curative treatments are available for late-stage metastatic prostate or rapidly progressing pancreatic cancers.

EXPERIMENTAL DESIGN Adenovirus type 5 mutants were created with deletions in the E1ACR2 region for tumor selectivity and/or the E1B19K gene for attenuated replication in vivo; all constructs retain the E3 genes intact. Cell-killing efficacy, replication, and cytotoxicity in combination with chemotherapeutics were investigated in normal cells (PrEC and NHBE), seven carcinoma cell lines, and human (PC3 and DU145) and murine (TRAMPC, CMT-64, and CMT-93) tumor models in vivo.

RESULTS The double-deleted AdDeltaDelta (DeltaE1ACR2 and DeltaE1B19K) mutant had high cell-killing activity in prostate, pancreatic, and lung carcinomas. Replication was similar to wild-type in all tumor cells and was attenuated in normal cells to levels less than the single-deleted AdDeltaCR2 mutant. AdDeltaDelta combined with the chemotherapeutics docetaxel and mitoxantrone resulted in synergistically enhanced cell killing and greatly improved antitumor efficacy in prostate xenografts in vivo. In murine immunocompetent in vivo models efficacy was greater for mutants with the E3B genes intact even in the absence of viral replication, indicating attenuated macrophage-dependent clearance.

CONCLUSIONS These data suggest that the novel oncolytic mutant AdDeltaDelta is a promising candidate for targeting of solid tumors specifically in combination with chemotherapeutics.

Zoopolis, vilda djurs suveränitet och predationsproblemet2018Independent thesis Basic level (degree of Bachelor), 10 credits / 15 HE creditsStudent thesis
    Abstract [sv]

The Problem of Predation in Zoopolis”.

Phosphorus Centers in π-conjugated Systems2012Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

Single-molecular electronics and organic material electronics are expanding research fields that ultimately aim for a vast variety of different applications, ranging from organic light-emitting diodes, to novel ways to improve the performance and decrease the size of electronics components. To achieve these goals, research has to be focused both on the development of functional molecules, but also on device fabrication. The work of this thesis is focused on the development of synthetic routes towards novel molecules for potential organic electronics applications, together with an investigation of their optical and electronic properties.

The first part of the thesis describes the synthesis of butadiyne-substituted and diacetylenic phosphaalkenes. Theoretical, spectroscopic and electrochemical techniques have been used to understand key steps during their synthesis, and to gain further information on the conjugative properties of their π-systems. A mechanism is proposed for the formation of the butadiyne-substituted and diacetylenic phosphaalkenes and it is shown that the phosphorus heteroatom is an intrinsic part of the π-conjugated system. The incorporation of the phosphorus heteroatom leads to decreased HOMO-LUMO gaps compared to all-carbon based reference compounds.

In the second part of the thesis, acetylenic phosphaalkenes are utilized for the preparation of phosphaalkene-substituted phospholes. A first step towards the exploration of the difference in reactivity of the σ2, λ3 phosphaalkene-P and the σ3, λ3 phosphole-P is presented as the oxidation of the compounds by sulfur proceeds selectively at the σ3, λ3–P. Spectroscopic and electrochemical investigations show that the phosphaalkene is an integral part of the compounds’ π-systems, and induces a HOMO-LUMO gap decrease compared to reference compounds that lack the P=C substituent.

The third part of this thesis presents an exploratory study concerning the suitability of metathesis reactions for the assembly of alkene-bridged phosphaalkenes.

    Abstract [en]

A series of C,C-diacetylenic phosphaalkenes 1b-e has been prepared from 1-chloropenta-1,2-dien-4-ynes 6b-e in a reaction with Mes*PCl2 (Mes* = 2,4,6-(Bu-1)(3)Ph) in the presence of LDA. Under identical conditions, isomeric butadiyne-substituted phosphaalkenes 2e-f can be obtained from 3-chloropenta-1,4-diyiies 5c-f. The title compounds represent rare examples of diethynylethenes in which a constituting methylene has been replaced by a phosphorus center. The formation of both isomers can be rationalized by a common pathway that involves isomeric allenyllithium species. Spectroscopic, electrochemical, and theoretical investigations show that the phosphorus heteroatoms are an intrinsic part of the compounds' pi-systems and lead to decreased HOMO-LUMO gaps compared to those in all-carbon-based reference compounds.

    Abstract [en]

The reactivity of bis-TMS-substituted C,C-diacetylenic phosphaalkene (A(2)PA) 1 in Sonogashira-Hagihara cross-coupling reactions has been examined. The selective and successive deprotection of the two silyl groups in 1 is enabled by the steric bulk of the Mes* group which renders the acetylene trans to Mes* more reactive and thereby facilitates selective and consecutive couplings with iodoarenes. In situ transformation of the TMS-protected acetylenes into Cu(I)acetylides is the key step in the synthetic sequence and enables the preparation of the first dimeric A(2)PA linked by a phenylene spacer. cis-trans Isomerization across the P = C bond is triggered by a tertiary amine and exclusively observed in the case of nitrophenyl-substituted A(2)PAs. The introduced aryl groups are integral parts of the entire p-system as evidenced by spectroscopic and electrochemical studies.

    Abstract [en]

Attempts to utilize C-ethylenic phosphaalkenes in metathesis reactions are discussed. Unprecedented reactivity is observed where the vinylphosphaalkene undergoes the first step of the catalytic cycle and cross-metathesis with the phenylmethylene moiety of Grubbs 2nd generation catalyst. However, homo-metathesis reaction to form 1,6-diphosphahexa-1,3,5-triene is not observed, presumably due to steric constraints.

Eutrophication has been a problem in aquatic systems for a long time. It is well known that the key nutrient to control the eutrophication process is phosphorus. Lately, it has been possible to reverse the process and recover lakes from eutrophication.

During a long time Lake Näsnaren in Katrineholms kommun, Södermanland, has been exposed of a large amount of phosphorus from the city's wastewater. The lake has a great value as a habitat of rare species but also a high recreation value for people living in Katrineholm and Södermanland. Lake Näsnaren is situated in a Natura 2000 area. This work aims to investigate Lake Näsnarens condition today, reasonable measures to reverse the eutrophication process and Lake Näsnaren's situation in the future.

Lake Näsnaren has today reached a very high level of phosphorus. Reducing the external load of phosphorus from fertilizers can reverse the eutrophication process. Increasing the depth also has a reversing effect.

If no measures are taken, it is likely that the lake overgrows. To reduce the lake's phosphorus values, more extreme measures have to be taken, like dredging or decreasing the load of phosphorus. However, Lake Näsnaren is shallow and reacts quickly to changes in the external load.

Purpose/Aim: To find power structures within the blog network.

Material/Method: Using keywords to find all available blog posts about the National Defence Radio Establishment from the blog search engine Twingly, and thereafter using the same blog search engine to find inlinks from other blogs, to those posts. The data is set into the context of the small-world networks models of Duncan J. Watts and rich-club models of Sergi Valverde och Ricard V. Sole.

Main results: 5183 unique blogs have written about the subject in 22779 blog posts to which 28128 inlinks from other blogposts are made. Just over one fifth of the blogs are linkted to each other in one big network, where the remaining blogs stand without any ingoing or outgoing links. The first bloggers are the one who continue to write for longer period of time and also the ones who attracts most inlinks. The blogosphere around this subject is highly connected, shows features as one would find in a small-world network, displays a power-law distribution for inlinks and is highly clustered around a few rich- club nodes.

High schools in Sweden, both public and private, have rapidly been adopting a computerbased learning platform. This means that each student is granted permission to lease a laptop from the school, which is intended to effectivize school work. However, moving the educational platform away from paper and pen and towards a digital platform poses certain risks concerning information security. This paper aims to examine, through a survey of 4 different high schools in Sweden, the education levels of students aged 16 to 19 concerning information security, and also how that education has been conducted. The study was divided into a survey of students, with 163 respondents, and interviews were conducted with 3 key members of 3 of the schools.

    Abstract [en]

This thesis aims to gain further understanding of the living conditions for widows in the cities of Vrindavan and Tiruchirappalli in India. Widows from all over India travel to Vrindavan, known as ”the city of widows”, when they have nowhere else to go, and Tiruchirappalli is home to a high number of farmer widows whom are being marginalized. This thesis seeks to problematize and shed light upon the alienation and difficulties these women face as a result of their widowhood, in relation to CEDAW. Based on three research questions it examines how the women’s living conditions could be understood in relation to Iris Marion Young’s theory about oppression; in which ways their living conditions become part of their identity because of their involuntary placement in the social group ”the widows”; and which implications this have on their rights. Young’s theory and Chandra Talpade Mohanty’s criticism regarding power and portrayal are being used to answer these questions. Information was provided through two independent fieldwork experiences conducted 2018 and 2019 at two NGO:s in said cities, both aiming to empower widows. The information consists of 14 interviews provided by participant-observation and a semi-structured interview method. The result shows that widows are being oppressed based on Young’s five criterions of oppression because of their involuntary placement in the social group ”the widows”, which in turn violates their rights as women and humans. However, the conclusion also states that related stigmatization continues to decrease, which could be a result of changing norms.

    Herbal melanin activates TLR4/NF-kappaB signaling pathway2009In: Phytomedicine, ISSN 0944-7113, E-ISSN 1618-095X, Vol. 16, no 5, p. 477-84Article in journal (Refereed)
    Abstract [en]

Expression of many pro-inflammatory cytokines is controlled by the NF-kappaB signaling pathway. NF-kappaB is induced by LPS through activation of TLR4. Melanins extracted from fungal, plant and human sources modulate cytokine production and activate NF-kappaB pathway. We showed that a herbal melanin (HM) from Nigella sativa L. modulates cytokine production and suggested it as a ligand for TLR4. In this study we investigated the possibility that the HM-induced cytokine production is via an NF-kappaB signaling pathway. We found that HM induced the degradation of IkappaBalpha, a key step in the activation of NF-kappaB. Moreover, addition of IkappaB kinase (IKK) specific inhibitors effectively inhibited the observed HM-induced production of IL-8 and IL-6 by TLR4-transfected HEK293 cells and THP-1 cells. Our results have also shown that HM induced cleavage of caspase 8, and that this cleavage was partially abrogated by IKK inhibitors. We suggest that HM can modulate the inflammatory response by inducing IL-8 and IL-6 production via TLR4-dependent activation of the NF-kappaB signaling pathway.

Cytokine-induced restoration of differentiation and cell cycle arrest in v-Myc transformed U-937 monoblasts correlates with reduced Myc activity2001In: Leukemia, ISSN 0887-6924, E-ISSN 1476-5551, Vol. 15, no 2, p. 217-227Article in journal (Refereed)
    Abstract [en]

Deregulated expression of the myc-family of oncogenes in hematopoietic and other cell types plays an important role in tumorigenesis, and results in increased proliferative potential and block of cellular differentiation. We have previously shown that IFN-gamma restores phorbol ester-induced differentiation and cell cycle arrest in v-myc transformed human U-937 monoblasts. To investigate whether other cytokine signals could also abrogate such a block, IL-1, IL-3, IL-4, IL-6, IL-7, IL-10, IL-11, LIF, oncostatin M, M-CSF, G-CSF and GM-CSF, and TGFbeta1, TNF-alpha, IFN-alpha were examined. We show that GM-CSF and IL-6, in combination with the phorbol ester 12-O-tetradecanoyl-phorbol acetate (TPA), restored differentiation and cell cycle arrest. In contrast, treatment by TGFbeta1 +/- TPA resulted in an efficient G1/G0 arrest, but did not appear to induce terminal differentiation. Restoration of differentiation and cell cycle arrest was accomplished despite maintained expression of the v-Myc protein. Our results show that the cytokine-induced signals reduced Myc-dependent transcription of an artificial target promoter/reporter gene construct, correlating in most, but not all, cases with decreased association of v- and c-Myc with its essential partner, Max. Thus, cytokine-induced signals may counteract the activity of deregulated Myc, and contribute to the normalization of differentiation, arrest in the G1/G0 phase of the cell cycle, or both.

    Abstract [en]

During World War II a number of actions were taken contrary to the traditional  Swedish freedom of the press. One of these actions was the establishment of “Statens Informationsstyrelse”, SIS,  the Gouvernment Board  of Information. SIS hade rhe task of managing and monitoring the Swedish press. It has subsequenly become known for the so-callade “grey notes”, cinfidential message to newspaper editors about what was not allowed to be published.

There were three newspapers in Gotlands during World War II. “Gotlänningen” represented the farmer´s party. The right-wing Gotlands Allehanda, whose principal shareholder C E Ekman was also the German consul, was often portrayed as very pro-nazi. The social demofratic “Gotlands Folkblad” was, on the other hand, potrayed as anti-nazi.

Based on the SIS archive, a review has been done about how SIS controlled the newspaperes and ruled through the greynotes and reprimands in case of transgressions.

Furthermore an analysis has been done on the different reprimands SIS took aginst the Gotland newspapers and if the reprimands were taken in response to the alleged anti-nazism of Gotlands Folkblad or the allegeds pro-nazism of Gotlands Allehanda.

The result show thar the vast majority of the articles SIS responded to in the Gotland newspapers were pure news articles. In no single case did SIS respond to a commentary article.

A GIS-based study of sites for decentralized composting and waste sorting stations in Kumasi, Ghana2011Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

Developing countries are facing a great challenge to collect, treat and dispose their waste in a more sustainable way. Today, most of the produced waste ends up on landfills, where they pose a great threat to the environment and human health. Kumasi, the second largest city in Ghana, faces the same issues as other cities in developing countries; waste management is run poorly and most waste ends up on the cities largest landfill, Dompoase, which will be full in a few years time. Issues such as low financial resources, bad urban planning and a growing population aggravate the implementation of a more sustainable development. Since most solid waste in developing countries is organic, composting is a good option towards a more sustainable waste management. There are numerous amounts of articles stating that decentralized composting is the best option for developed countries. Decentralized compost facilities are less costly to install and maintain, they require less technology and decrease the cost of waste transportation. Transportation poses a large expense when it comes  to waste management in developing countries. There is an advantage in integrating a more sustainable waste management in to the existing waste infrastructure since cities often are densely built up and there is a shortage of land. Therefore this thesis has investigated the possibilities of having waste sorting stations and decentralized compost facilities at public dumping sites in the Bantama area, one of ten sub--‐metros in Kumasi. Based on literature and observations during field visits in the Bantama area, a classification scheme was developed. The classification scheme was designed to simplify identification and differentiation of the sites. It contains thirteen criteria to consider when planning for organic waste sorting stations and decentralized compost facilities. Suitable sites for sorting stations and decentralized compost facilities were selected by a SQL analyzes in a Geographic Information System (GIS). The analyzes was based on the classification scheme. The result shows that of twenty--‐one public dumpingsites, seven were suitable as sorting stations and two sites were suitable for a decentralized compost facility. The expectation is that in due time and with infrastructure improvements, more than only seven communal sites can become sorting stations. When it comes to sites for composting the result reveals an issue in many cities  there is just not enough land to build as many decentralized facilities as would be necessary to recycle all organic waste. However, due to the advantages of having decentralized facilities, the recommendation is that Kumasi should start with a decentralized approach and as finance and technological skills exists the organic waste management can extend to also include larger facilities. Further this thesis shows the advantage of having decentralized composting when it comes totransportation costs. A transport analyzes was done in a GIS,  which showed the difference intransportation distances between having decentralized and centralized composting. The result showed that waste recycled close to its source, i.e. having decentralized waste management, is transported shorter distance compared to having centralized composting and  thus saving on cost for transportation.

Why has a ‘little Sweden’ emerged in Brazil?: A study of Swedish direct investments in Brazil2007Independent thesis Advanced level (degree of Master (One Year)), 10 credits / 15 HE creditsStudent thesis
    Abstract [en]

Not long ago, Brazil was a country with an unstable economic system; inflation and interest rates were high and the political system was corrupt and complex. Still Brazil has for a long time been a successful market for Swedish companies. In São Paulo, the financial center of Brazil, a “little Sweden” has emerged which is considered to be the largest center for Swedish industry outside of Sweden.

The purpose of this thesis is to examine why a ‘little Sweden’ has emerged in Brazil as well as examining what importance different economical factors have had for Swedish foreign direct investments in Brazil. Theories regarding foreign direct investments as well as other business theories have been used in the research. To get a deep understanding of the subject, four interviews have been made with organizations involved in helping Swedish companies in Brazil.

The research has shown that the most important factors in the creation of the ‘little Sweden’ has been; access to production factors such as natural resources, the competitiveness of the home market together with the size of Brazil’s market, the financial capacity of the Swedish companies, the fact that Swedish companies could offer products and services that suited the demands of Brazil, as well as the fact that the Swedish entrepreneurs had a long term thinking and a great optimistic visions for the future.

    Abstract [en]

This thesis concerns an energy audit of LKAB Logistics. According toswedish law a company the size of LKAB are required to make an energyaudit every four year. LKAB Logistics mainly transports refined ironore products between mines and harbours along the train track"Malmbanan". They also store iron ore products and load boats in theharbours in Narvik and Luleå. These kinds of operations have a highdemand for energy. This demand is met mostly with electricity. However, oil, gas, diesel and district heating are also consumed tomeet the energy demand. In total LKAB Logistics consume around 191GWh, where roughly 75% is of electricity. With such high demand, savings around 1-2% makes a significant difference, and there areopportunities and incentives to improve energy efficiency.Implementing staff that are responsible for energy work, makinginvestments in new electrical motors and more energy-efficient windowsare a few examples. Higher priority on energy saving and continuedprogress for a more efficient work place will most likely benefit thecompany's environmental goals and economy.

Skiftet i Enåker vid mitten av 1800-talet: En undersökning om laga skiftets påverkan på äng och åker2018Student paper second term, 5 credits / 7,5 HE creditsStudent thesis
    Abstract [en]

The purpose of this essay is to examine the changes in the agricultural sector during the mid-nineteenth century. It was during this period "laga skiftet" was enacted, which was a law aimed at improving the efficiency of farms by changing the land structure within the collective of farms called "sock". The principal method proposed by "laga skiftet" was to change land from the pasture grounds to the field crop areas. Through meticulous study of original documents of the period, statistics were compiled and presented to show the effects on Enåker’s land structure. The result is conclusive and as intended by the law change, but minor anomalies exist. This is due to the unexplainable relation between the growth in field crop areas and the shrinking of pasture grounds during certain periods, leaving, in some cases, a large amount of hectare’s origins unexplained. Efforts are made to analyse and address this mystery, citing factors such as the use of forest areas and the marsh areas as previously untapped land. Other explanations are given for this disparity including macro-economic, technological and demand side factors. The conclusion finds the law change successful in achieving its goals but recognises other factors also contributed to the changing agricultural landscape during this time. With greater further research into this topic, building upon the groundwork in this text, answers to these anomalies may be unearthed and a wider understanding of the effects and merits of "laga skiftet” will become clear.

Hur överförs tyst kunskap?: En studie av kunskapsintensiva företags tillämpade metoder2013Independent thesis Basic level (degree of Bachelor), 10 credits / 15 HE creditsStudent thesis
    Abstract [sv]

Validation of a species-optimized enzyme-linked immunosorbent assay for determination of serum concentrations of insulin in dogs2011In: Veterinary clinical pathology, ISSN 0275-6382, E-ISSN 1939-165X, Vol. 40, no 1, p. 66-73Article in journal (Refereed)
    Abstract [en]

BACKGROUND: Measurement of canine serum insulin has relied on methods developed to measure human insulin. A species-optimized test for measurement of serum insulin in dogs is now commercially available.

OBJECTIVE: The purpose of this study was to validate the canine ELISA for determination of serum insulin concentration in dogs.

METHODS: Precision was determined by evaluating intra- and interassay coefficient of variation (CV), and accuracy was determined by dilution and spike recovery studies. A method comparison study with samples from 34 clinically healthy dogs and 73 dogs examined for various illnesses and disorders ("patients") was performed using the canine ELISA and an ELISA for human insulin. Biologic relevance of the canine assay was evaluated by measuring insulin in samples collected from 8 healthy dogs after administration of glucagon. A stability study was preformed with 6 samples stored at 20°C, 4-8°C, and -20°C.

RESULTS: For the canine ELISA, intra- and interassay CVs were 4.3-7.8% and 4.4-7.7%, respectively. Mean recovery after dilution was 99% and recovery after spiking with porcine insulin was 116%. The canine and human ELISAs correlated well (r(2) =.94 for healthy dogs, r(2) =.88 for patient samples). After glucagon injection serum insulin concentrations increased significantly in 8 dogs. Insulin was stable for 30 days in 6 serum samples stored at -20°C and in most samples for 8 days at 4-8°C. Insulin was stable for <3 days at room temperature (20°C).

CONCLUSIONS: The new canine serum insulin ELISA had good precision and accuracy and correlated well with the previously used assay.

An Analysis and Interpretation of the Geoscience BC Trek Project Till Geochemical and Mineralogical Data to Determine the Surficial Geochemical Expression of Bedrock Au-Ag-Cu-Mo Mineralization on the Nechako Plateau, British Columbia, Canada.2016Independent thesis Basic level (degree of Bachelor), 10 credits / 15 HE creditsStudent thesis
    Abstract [en]

The mineral exploration in the interior of British Columbia is challenged by an extensive coverage of glacial sediments. The Geoscience British Columbia (GBC) Targeting Resources through Exploration and Knowledge (TREK) Survey has since 2013 collected till samples in the northern interior plateau of British Columbia, covering an area of approximately 25,000km2. The TREK area is largely covered by Miocene to Pleistocene age volcanic rocks that are underlain by older volcanic, sedimentary, and intrusive rocks of the Stikine Terrane, and also to a smaller extent in the east by rocks of the Cache Creek Terrane. Previous surficial mapping to aid mineral exploration has determined that the Pleistocene ice sheet advances have been mainly to the east-northeast. The British Columbia Geological Survey (BCGS) MINFILE database includes five developed prospects, seven prospects and 39 mineral showings in the TREK Project area. Styles of mineralization include late Cretaceous and Eocene epithermal Au and Ag deposits (e.g., Blackwater-Davidson, Capoose, Wolf, and 3T’s), lower Cretaceous sediment-hosted Carlin-type deposits (e.g. Bob) and Jurassic to Eocene porphyry Cu and Mo deposits (e.g., Endako and Chu). This report analyzes and interprets till digital data from three TREK Geoscience BC reports and one BCGS Open File to determine if there is a mineralogical and geochemical signature in the till of buried bedrock mineralization. Through statistical analysis, including scatter plots, probability plots, and box plots, populations of key ore indicators and pathfinder elements are compared, thresholds are established and anomalous samples queried. Spatial plots of samples with known pathfinder elements e.g. Au, Ag, Cu, and Mo values above established thresholds are created and compared with heavy mineral till concentrates such as pyrite and cinnabar. Major oxide element ratios from till sample whole rock analysis were also analyzed to identify hydrothermal alteration associated with the mineralization. The analysis has demonstrated that anomalous values reflect known bedrock mineralization and that there are two areas within the study area that do not appear to be related to already known mineral occurrences.

On Conditions of Swedish Women’s Sexual Well-Being: An Epidemiological Approach2005Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

Objectives: This descriptive epidemiological dissertation aims to identify conditions of Swedish women’s sexual well-being. The focus is on the relationship between their idiosyncratically reported levels, during the last 12 months, of 5 sexual functions/dysfunctions per se and distressing and their socio-psychological situation, including aspects of their sexual history. Levels of sexual functions/dysfunctions are also related to levels of sexual satisfaction and to other aspects of life satisfaction.

Methods: Data on a randomized cross-sectional national sample of 1335 women aged 18-74 (59% of target sample) were gathered in 1996 using a combination of structured interviews and questionnaires/checklists. Analyses were performed for the total sample or for sub-samples aged 18-65 years. In 3 of the 4 dissertational articles, trichotomies of a 6-grade scale characterizing level of sexual dysfunctions into No/Mild/Manifest dysfunction were used.

Main results: Mild sexual dysfunctions were, generally, much more common than were manifest, and dysfunctional distress was considerably less common than were dysfunctions per se. All dysfunctions, and in particular orgasmic dysfunction, were closely associated with level of sexual well-being. Four factors independently pair-wise linking levels of dysfunctions per se with levels of distressful dysfunction were identified. These were Sexual interest/Desire, Genital function (lubrication and dyspareunia), Orgasm, Vaginismus. Three of these (not vaginismus) were powerful classificators of gross level of sexual well-being. Many of socio-demographic and socio-psychological contextual life-conditions were significantly associated with the different sexual functions/dysfunctions. However, the most prominent contextual variables were satisfaction with partner relationship and partner’s levels of sexual functions.

In conclusion, many different socio-psychological aspects must be taken into account to optimize treatment modalities and resources when dealing with women’s sexual dysfunction in order to secure a good level of sexual well-being.
