Angiogenesis - formation of new capillary blood vessels - is essential during development and physiological conditions that require angiogenesis, such as wound healing and the menstrual cycle. Prolonged and excessive angiogenesis has been implicated in a number of pathological processes, for instance rheumatoid arthritis, retinopathy and tumor growth. Preventing angiogenesis as a treatment strategy for diseases characterized by excessive angiogenesis is therefore of interest. To date, five anti-angiogenic drugs have been approved by the FDA, for the treatment of cancer and an ocular disorder with exaggerated neovascularization.The aim of the proposed research is to establish new and effective treatment strategies to prevent angiogenesis under pathological conditions, with a special focus on cancer. Histidine-rich glycoprotein (HRG) has been identified as an inhibitor of angiogenesis and tumor growth. We are addressing the mechanism/s behind the negative regulation of angiogenesis by HRG, with a special focus on the role of platelets in this process.Moreover, we are establishing a novel strategy for therapeutic vaccination against tumor vascular antigens as a cancer therapy. The unique aspect of the vaccination project is the combination of an effective therapeutic vaccination technology with antigens specific for tumor vessels.
