Molecular interactions beyond induced fit and affinity analysis: Relevance for regulation of biological processes and drug discovery

OVERALL AIMS: To develop the basic science of molecular recognition and obtain a fundamental understanding of the dynamic characteristics of molecular interactions. STRATEGY: To use a biochemical approach based on time-resolved interaction analysis using SPR-biosensor technology, primarily complemented by structural and computational methods. Studies will involve simple protein-ligand systems and complex molecular systems of relevance for basic biological processes and drug discovery. PREVIOUS RESULTS: We have established biosensor technology for detailed studies of molecular interactions and studied the kinetics, mechanisms, dynamics, thermodynamics and chemodynamics of a variety of molecular systems. This has contributed to a better understanding of molecular interactions, especially concerning kinetics and mechanistic complexities, such as conformational dynamics. It has had a significant impact on enzymology and drug discovery. Here we will continue our work on the discovery of drugs against HIV, HCV, Alzheimer´s disease and other neurological diseases, while exploiting our expertise in interaction analysis of membrane bound proteins for investigation of the replication complex of HCV and other membrane associated enzyme systems. SIGNIFICANCE: The project will contribute to the field of molecular recognition with relevance for many cellular and physiological processes, and their regulation, as well as to a more efficient development of better drugs.
