Title [sv]
Betydelsen av elektrostatisk växelverkan i komplex av polyelektrolytnätverk och proteiner
Title [en]
Importance of electrostatic interaction in complexes between polyelectrolyte networks and proteins
Abstract [sv]
The project deals with the interaction between charge polymer networks and oppositely charge macroions. Previous results indicate that core/shell phase separation is often observed when proteins bind to microgels, and that the formation of dens shells can strongly reduce binding and deswelling rates. We aim at a detailed understanding of how electrostatic interactions influence on the shell formation. In particular we will investigate how variations of the protein and network charge densities influence on the appearance of shells, the swelling of the shell structures, and the relation between the compactness of the shell structure and the transport of protein in it. For this purpose we will use as models for proteins two types of model macroions: dendrimers and quantum dots. These allow for systematic variations of charge without strong interference of other interactions. The unique photophysical properties of quantum dots will make it possible to determine concentration profile in the gels and the total amount taken up by a single gel in a laser scanning confocal microscope experiment, which is the main technique that will be employed. The results from the project will have implications to the use of polymer gels as delivery systems for protein drugs.
