A tumor does not consist of a homogenous population of cancer cells. Therefore, to understand cancer, the tumor microenvironment and the interplay between the different cell types present in the tumor has to be taken into account, and how this interplay regulates the growth and survival of the cancer cells. The aim with this project is to simultaneously analyze the activity status of multiple signal pathways at a single cell level. To achieve this we will take advantage of the in situ PLA method that was developed in our group, for monitoring protein interactions and post-translational modifications in tissue sections. However, to acquire a coherent view on the cellular activity status of each individual cell within a tumor section we will need to use a highly multiplexed in situ PLA that targets several signaling pathways, at different nodes in the cascade. This will provide information on what pathways are active in cancer cells, and to what extent this varies depending on positioning within the tumor, and in addition it will reveal how the surrounding non-malignant cells in the tumor microenvironment are affected by the cancer cells. Hopefully, this knowledge will also enable better diagnostics, improved prediction on response to therapy and may possibly also act as an incitement to develop novel drugs that can modify the microenvironment, thereby reducing cancer growth and its ability to metastasize.
