Abstract [en]
In this proof of concept project, arising from our ongoing VR project PharmaPLA, we will further characterize a new method we have developed to investigate specific and nonspecific target engagement by DNA modified drug candidates. The attached DNA strands enable highly sensitive detection of target binding in protein arrays, cell preparations, and tissue section, by using our portfolio of molecular genetic techniques, using techniques such as in situ proximity ligation or unfolding assays with localized signal amplification via rolling circle amplification, and the recently developed super RCA technique. The project will involve further characterization of the molecular procedures and their suitability for evaluating new drug candidates; and we will investigate our IP position and optimal paths towards a commercial product as a kit or service.The project will be conducted at Uppsala University, but with assistance from our spin-out Olink Bioscience. It addresses a central need in the development of new therapeutic entities, and potential results include both more efficient drug development and establishment of a valuable product line at Olink or a spin-out, using our unique molecular tool kit.
