The effect of the gene Zmynd15 on the Caco2 colon cancer cell line

Colorectal cancer is one of the most occurring cancer forms, and accounts for 9,7 % of all the cancer cases and makes up for 8,5 % of death cases worldwide. Common treatments for colon cancer are chemotherapy and tumor removal through surgery. This study aims to examine potential roles of the gene Zmynd15 in colon cancer where it is unstudied. A known function of Zmynd15 is that the protein acts as a transcription repressor in the interaction with histone deacetylase which removes acetyl groups from histones, thus giving histones a positive charge, causing the DNA to bind easier to the histones, which suppresses transcription. Methods used in this study includes RNA inhibition by siRNA, flow cytometry that examines the effect of Zmynd15 on the cell cycle phases, fluorometric microculture cytotoxicity assay to measure chemotherapeutic resistance, western blot to analyze the effect on endosomal protein expression and an oxygraph to analyze the metabolic effects. The results showed that the gene had no effect on either the cell cycle or most of the drugs in the chemotherapeutic resistance analysis, but some concentrations need to be further examined due to differences observed. The endosomal markers, except for EEA1, could not be detected in the western blot. No difference in oxygen consumption was obtained. This study showed that the protein did not contribute to tumor growth or suppression, metabolic pathways or contributed to drug resistance and that more research is needed to understand the role of Zmynd15.
