A validation of ultrasound with histology for measurement of abdominal aortic aneurysm in mice

Abstract [en]

Abdominal aortic aneurysm is a vascular disease that has increased in the Western world, with a prevalence of circa 2 %. Aneurysm implies that the aorta grows beyond its normal size, until it ruptures, and the patient dies. To be defined as an aneurysm, the abdominal aorta needs a diameter of at least 30 mm. Right now the most common way to treat aneurysm is with surgery. Because of the risks associated with surgery, a more preventative approach is needed.    

  The purpose of this report was to validate two different methods, ultrasound with histology, to find out if the medicine Bosutinib had any effect on the growth of aneurysm in mice. Aneurysm was induced by researchers giving the mice angiotensin II through a mini-osmotic pump. After that, the mice were treated with Bosutinib trough an oral gavage for 22 days. The mice were split into three groups, 10 mg/kg, 100 mg/kg and a control group. Samples from the abdominal aorta were collected from the mice, and the samples were cryosectioned and the sections stained with a hematoxylin-eosin staining. Pictures were taken of the sections. From the pictures the area and circumference were measured, and diameter were calculated from these measurements.    

  Because of different problems during the practical work, it was only possible to collect data from eight mice. From these data it was possible to see a correlation that treatment with Bosutinib also led to a smaller aortic diameter using both methods.
